Morphology and Structure

Trichomes are straight, loose, immobile or weakly motile, up to 15 µm long, and consist of up to 10 cells. Vegetative cells are cylindrical, isodiametric, sometimes barrel-shaped, light blue-green, homogeneous, non-granular, 1.8–6.8 × 2.3–4.3 µm. Heterocysts are terminal, unipolar, spherical or elongated, yellow-green, 3.0–5.9 × 2.3–3.2 µm. Akinetes are predominantly cylindrical or oval, large, light brown or yellow, granular, 5.1–7.7 × 2.6–3.4 µm, adjacent to heterocysts; sometimes two akinetes occur at one end of the trichome. Reproduction occurs by binary cell division in one plane. On solid medium, the strain forms mucilaginous green colonies that later become brownish. In old cultures, heterocysts and akinetes are observed on days 10–15 of cultivation.

Cultivation

  • Nutrient media: Allen's, BG-11, Bold's (liquid and agarized)
  • Temperature: 25–30 °C
  • Light intensity: 30–40 µmol photons/m²/s
  • Photoperiod: 16:8 h (light/dark)
  • pH: 7.0

Physiological Characteristics

Mesophilic heterocystous cyanobacterium capable of forming terminal heterocysts and akinetes. The strain has nitrogen-fixing potential; in the acetylene reduction assay, its activity was 1.36 ± 0.05 nmol ethylene mg⁻¹ dry weight h⁻¹.

Long-term Preservation Conditions

The strain is stored on agarized BG-11 medium at 12–14 °C under low illumination of 4–6 µmol photons/m²/s. Maximum storage period without subculturing: 2–3 months.

Genetic Information

A fragment of the 16S rRNA gene was amplified. Based on morphological and phylogenetic analysis, the strain was identified as Cylindrospermum badium J-8. GenBank accession number: MZ079357.

Field of Application

The strain is used in experimental studies. It is promising for the development of biofertilizers, bioremediation, and algal-cyanobacterial biofilm formation.
