- Genetic features: Wild type (WT)
- Culture condition: Axenic
Morphology and Structure
Trichomes are filamentous and surrounded by a mucilaginous sheath; active motility is not observed, but weak oscillation of trichome tips is noted. Vegetative cells are blue-green, 2.6–5.0 µm wide; cell length is 2–3 times greater than cell width. Heterocysts are absent. Akinetes are absent. Reproduction occurs by binary cell division in one plane. In liquid medium, the strain forms a blue-green slightly mucilaginous suspension with an amorphous sediment that is easily resuspended by shaking. On agarized Gromov medium, it forms blue-green spherical clusters of trichomes. The culture develops independently of season.
Cultivation
- Nutrient media: Gromov's, Allen's, BG-11 (liquid and agarized)
- Temperature: 25–28 °C
- Light intensity: 30–40 µmol photons/m²/s
- Photoperiod: continuous illumination
- pH: 7.5–8.0
Physiological Characteristics
Mesophilic homocytous filamentous cyanobacterium. It does not form heterocysts or akinetes. Under the experimental conditions, nitrogen-fixing activity was not detected, which corresponds to the absence of heterocysts in this morphotype.
Long-term Preservation Conditions
stored on agarized BG-11 medium at 12–14 °C under low illumination of 4–6 µmol photons/m²/s. Maximum storage period without subculturing: 2–3 months.
Genetic Information
A fragment of the 16S rRNA gene was amplified. Based on morphological and phylogenetic analysis, the strain was identified as Oscillatoria brevis SH-12. GenBank accession number: MZ090011.
Field of Application
The strain is used in experimental studies. It is promising for biotreatment, bioremediation, and algal-cyanobacterial biofilm formation. It may also be used as a phototrophic model culture among non-heterocystous filamentous cyanobacteria.
